Say I wanted to get from Littleton to Vail on a CRF450X with trail gearing...

Is there any way to avoid highways? Or am I shit outta luck?

The more dirt the better, sub 50 MPH roads would be fine.

How much time do you have and how difficult can the route be?

I have all day and a 3.2g tank. (140ish mile range I'd guess)

It can be plenty difficult.
